چکیده

The importance of quantitative studies of the specificities of purine-metabolizing enzymes are discussed. These data not only enable one to make predictions concerning the metabolic disposition and effectiveness of a drug in vivo, but they also help to identify its loci of action. They may also serve as a basis for understanding the selective toxicity of a compound and aid in the design of new antimetabolites.

Substrate specificity of human recombinant mitochondrial deoxyguanosine kinase with cytostatic and antiviral purine and pyrimidine analogs.

Deoxyguanosine kinase (dGK) is an enzyme responsible for the phosphorylation of purine deoxynucleosides in mitochondria of mammalian cells. Its role in activation of pharmacologically used nucleoside analogs is not well understood, because of the low levels of dGK found in tissue extracts and its inactivation during purification. The cDNA for dGK was recently cloned and expressed in Escherichia...

Selective abolishment of pyrimidine nucleoside kinase activity of herpes simplex virus type 1 thymidine kinase by mutation of alanine-167 to tyrosine.

Herpes simplex virus type 1 (HSV-1) encodes a thymidine kinase (TK) that markedly differs from mammalian nucleoside kinases in terms of substrate specificity. It recognizes both pyrimidine 2'-deoxynucleosides and a variety of purine nucleoside analogs. Based on a computer modeling study and in an attempt to modify this specificity, an HSV-1 TK mutant enzyme containing an alanine-to-tyrosine mut...

Purine salvage in two halophilic archaea: characterization of salvage pathways and isolation of mutants resistant to purine analogs.

In exponentially growing cultures of the extreme halophile Halobacterium halobium and the moderate halophile Haloferax volcanii, growth characteristics including intracellular protein levels, RNA content, and nucleotide pool sizes were analyzed. This is the first report on pool sizes of nucleoside triphosphates, NAD, and PRPP (5-phosphoribosyl-alpha-1-pyrophosphate) in archaea. The presence of ...

Alternate Substrates and In hi bitors of 1 -Aminocyclopropane-I -carboxylic Acid Synthase

Structural analogs of (-)-S-adenosyl+methionine (SAM), in which the heterocyclic base was modified, were used to initiate studies to elucidate the active site conformation of the enzyme l-aminocyclopropane-I-carboxylic acid (ACC) synthase, which was partially purified from Lycopersicon esculentum (tomato). These potential substrate analogs were screened for activity both as substrates and/or as...

Kinetic study of peroxidase enzyme in the presence of purine alkaloids

Peroxidase catalyzes different oxidation of substrates using hydrogen peroxide, a reactive oxygen specie (ROS). ROS, at low concentrations, act as messenger to regulate intracellular signaling, whereas, at high concentrations, they can overcome the immune system by creating oxidative stress. Some common beverages such as coffee, tea and soft drinks contain high levels of xanthine alkaloids incl...
